Sending an e-mail is by far the most effective way to correct the mistake.
Just as if you placed an order with Amazon, the purchase runs through immediately, meaning you can't really add to the order after it's placed. Unfortunately, we've all been spoiled thanks to BrickLink, which allows us to continue to add to our order - the difference is that the orders aren't pre-paid and are invoiced after the fact.
It usually takes us 24 hrs to get to your order, and most of the time we'll read customer inquiries prior to packing.
